The long-term impact of elevated concentration of CO2 on assimilation activity of sun-exposed (E) versus shaded (S) foliage was investigated in a Norway spruce stand [Picea abies (L.) Karst, age 14 years] after three years of cultivation in two domes with adjustable windows (DAW). One DAW was supplied with ambient air [AC, ca. 350 µmol(CO2) mol–1) and the second with elevated CO2 concentration [EC = AC plus 350 µmol(CO2) mol–1]. The pronounced vertical profile of the photosynthetic photon flux density (PPFD) led to the typical differentiation of the photosynthetic apparatus between the shaded and sun needles. Namely, photon-saturated values of maximal net photosynthetic rate (P Nmax) and apparent quantum yield () were significantly higher/lower for E-needles as compared with the S-ones. The prolonged exposure to EC was responsible for the apparent assimilatory activity stimulation observed mainly in deeply shaded needles. The degree of this stimulation decreases in the order: S-needles dense part > S-needles sparse part > E-needles dense part > E-needles sparse part. In exposed needles some signals on a manifestation of the acclimation depression of the photosynthetic activity were found. The long-term effect of EC was responsible for the decrease of nitrogen content of needles and for its smoother gradient between E- and S-needles. The obtained results indicate that the E- and S-foliage respond differently to the long-term impact of EC.  相似文献

Hepatocyte growth factor (HGF) is a mesenchymal-derived cytokine. It exerts in vitro a motogenic effect on various target cells, which is displayed either by cell scattering, locomotion, and migration during the wound repair process of cultured cells, or invasiveness through the extracellular matrix. Although it is known that HGF influences the motogenic effect of endothelial cells, the precise effects of HGF during angiogenesis are still poorly understood. To identify genes regulated via HGF signaling in HUVECs, we used the differential display polymerase chain reaction. In this study, thymosin beta4 was found to be differentially expressed in HGF-treated HUVECs compared with control. Data from HPLC profile and induction of MMPs indicate that HGF may affect the biological behavior of HUVECs through a combination of the direct effects of HGF itself and indirect effects mediated via induction of thymosin beta4 in vitro.  相似文献

In addition to proton-pumping complex I, plant mitochondria contain several type II NAD(P)H dehydrogenases in the electron transport chain. The extra enzymes allow the nonenergy-conserving electron transfer from cytoplasmic and matrix NAD(P)H to ubiquinone. We have investigated the type II NAD(P)H dehydrogenase gene families in Arabidopsis. This model plant contains two and four genes closely related to potato (Solanum tuberosum) genes nda1 and ndb1, respectively. A novel homolog, termed ndc1, with a lower but significant similarity to potato nda1 and ndb1, is also present. All genes are expressed in several organs of the plant. Among the nda genes, expression of nda1, but not nda2, is dependent on light and circadian regulation, suggesting separate roles in photosynthesis-associated and other respiratory NADH oxidation. Genes from all three gene families encode proteins exclusively targeted to mitochondria, as revealed by expression of green fluorescent fusion proteins and by western blotting of fractionated cells. Phylogenetic analysis indicates that ndc1 affiliates with cyanobacterial type II NADH dehydrogenase genes, suggesting that this gene entered the eukaryotic cell via the chloroplast progenitor. The ndc1 should then have been transferred to the nucleus and acquired a signal for mitochondrial targeting of the protein product. Although they are of different origin, the nda, ndb, and ndc genes carry an identical intron position.  相似文献

Diphyllobothrium pacificum has been reported as a human parasite from coprolites and skeletons in Peru and Chile. Our analysis of Chinchorro mummies from Chile provides the oldest evidence of D. pacificum directly associated with human mummies. These mummies date between 4,000 and 5,000 years ago. The basis for our diagnosis is presented. We find that the size of the eggs in the mummies is smaller than other discoveries of D. pacificum. We suggest that this is due to the peculiar circumstances of preservation of parasite eggs within mummies and the release of immature eggs into the intestinal tract as the tapeworms decompose after the death of the host. This information is important to consider when making diagnoses from mummies.  相似文献

This study presents a small-scale polymerization of high molecular weight methyl methacrylate/n-butyl acrylate (MMA/n-BA) colloidal particles that are synthesized in an aqueous environment in the presence of phospholipid hydrogenated soybean phosphatidylcholine (HSPC) molecules that also serve as the particle stabilizing agents. When such particles coalesce to form polymeric films, they release phospholipids, which, in turn, form organized structures near the film-air (F-A) interface. Diffusion and mobility of phospholipid molecules are affected not only by their compatibility with colloidal particles but also by electrolyte environments of colloidal dispersions. When Na(+), K(+), and Ca(2+) counterions are added to MMA/n-BA aqueous colloidal dispersions stabilized with HSPC, and such films are coalesced, different degrees of diffusion of HSPC to the F-A interface exist, depending on the counterion, and conformational changes of HSPC result. For example, in the presence of Ca(2+), HSPC molecules collapse entropically to form random surface layers, as opposed to smaller Na(+) and K(+), which force amphiphilic HSPC ends to align preferentially parallel to the film surface. These studies show that it is possible to design stimuli-response colloidal systems triggered by chemical environments of active molecules on colloidal polymer particles.  相似文献

The dynamics of the terrestrial ecosystems depend on interactions between a number of biogeochemical cycles (i.e. carbon, nutrient, and hydrological cycles) that may be modified by human actions. Conversely, terrestrial ecosystems are important components of these cycles that create the sources and sinks of important greenhouse gases (e.g. carbon dioxide, methane, nitrous oxide). Especially, carbon is exchanged naturally among these ecosystems and the atmosphere through photosynthesis, respiration, decomposition, and combustion processes. Continuous increase of atmospheric carbon dioxide (CO2) concentration has led to extensive research over the last two decades, during which more then 1 400 scientific papers describing impacts of elevated [CO2] (EC) on photosynthesis have been published. However, the degree of response is very variable, depending on species, growing conditions, mineral nutrition, and duration of CO2 enrichment. In this review, I have summarised the major physiological responses of plants, in particular of trees, to EC including molecular and primary, especially photosynthetic, physiological responses. Likewise, secondary (photosynthate translocation and plant water status) and tertiary whole plant responses including also plant to plant competition are shown.  相似文献

Gas exchange and chlorophyll (Chl) fluorescence were measured on young mature leaves of rose plants (Rosa hybrida cvs. First Red and Twingo) grown in two near-to-tight greenhouses, one under control ambient CO2 concentration, AC (355 µmol mol–1) and one under CO2 enrichment, EC (700 µmol mol–1), during four flushes from late June to early November. Supply of water and mineral elements was non-limiting while temperature was allowed to rise freely during daytime. Leaf diffusive conductance was not significantly reduced at EC but net photosynthetic rate increased by more than 100 %. Although the concentration of total non-structural saccharides was substantially higher in the leaves from the greenhouse with EC, PS2 (quantum efficiency of radiation use) around noon was not significantly reduced at EC indicating that there was no down-regulation of electron transport. Moreover, CO2 enrichment did not cause any increase in the risk of photo-damage, as estimated by the 1 – qP parameter. Non-photochemical quenching was even higher in the greenhouse with EC during the two summer flushes, when temperature and photosynthetic photon flux density (PPFD) were the highest. Hence rose photosynthesis benefits strongly from high concentrations of atmospheric CO2 at both high and moderate temperatures and PPFD.  相似文献

The acclimation depression of capacity of photon utilisation in photochemical reactions of photosystem 2 (PS2) can develop already after three months of cultivation of the Norway spruces (Picea abies [L.] Karst.) under elevated concentrations of CO2 (i.e., ambient, AC, + 350 µmol(CO2) mol–1 = EC) in glass domes with adjustable windows. To examine the role that duration of EC plays in acclimation response, we determined pigment contents, rate of photosynthesis, and parameters of chlorophyll a fluorescence for sun and shade needles after three seasons of EC exposure. We found responses of shaded and exposed needles to EC. Whereas the shaded needles still profited from the EC and revealed stimulated electron transport, for the exposed needles the stimulation of both electron transport activity and irradiance saturated rate of CO2 assimilation (P Nmax) under EC already disappeared. No signs of the PS2 impairment were observed as judged from high values of potential quantum yield of PS2 photochemistry (FV/FM) and uniform kinetics of QA reoxidation for all variants. Therefore, the long-term acclimation of the sun-exposed needles to EC is not necessarily accompanied with the damage to the PS2 reaction centres. The eco-physiological significance of the reported differentiation between the responses of shaded and sun exposed needles to prolonged EC may be in changed contribution of the upper and lower crown layers to the production activity of the tree. Whereas for the AC spruces, P Nmax of shaded needles was only less than 25 % compared to exposed ones, for the EC spruces the P Nmax of shaded needles reached nearly 40 % of that estimated for the exposed ones. Thus, the lower shaded part of the crown may become an effective consumer of CO2.  相似文献

Ubiquitin conjugation during endoplasmic-reticulum-associated degradation (ERAD) depends on the activity of Ubc7. Here we show that Ubc1 acts as a further ubiquitin-conjugating enzyme in this pathway. Absence of both enzymes results in marked stabilization of an ERAD substrate and induction of the unfolded-protein response (UPR). Furthermore, basic ERAD activity is sufficient to eliminate unfolded proteins under normal conditions. However, when stress is applied, the UPR is required to increase ERAD activity. We thus demonstrate, for the first time, a regulatory loop between ERAD and the UPR, which is essential for normal growth of yeast cells.  相似文献
